The road is expected to be closed in both directions until at least 5.30pm after the collision.
Kent Police attended the crash on the northbound carriageway near Pembury at 12.22pm.
Early reports say the collision involved a man driving a car and a van, also driven by a male.
The driver of the car was taken to Royal London Hospital in Whitechapel and a female passenger was being taken to King's College Hospital in London.
The van driver was taken to Tunbridge Wells Hospital.
Police confirmed all three casualties were conscious when they attended the scene.
Traffic is now being diverted onto the A26 via Vauxhall Lane and North Farm Retail Park but motorists are being urged to avoid the area if possible.
